This study describes a rapid purification of insulin-like growth factor-I from chicken serum and the immunological, biological and receptor binding activity of the peptide. It was purified after initial extraction, by cation exchange chromatography, hydrophobic interaction chromatography and reverse phase chromatography up to 1.4 x 10(6)-fold with an overall yield ranging from 10-30%. The N-terminal amino acid sequence was the same as predicted from the nucleotide sequence of a chicken IGF-I cDNA and the partial sequence obtained from a previously reported purification. The material was both immunologically and biologically active. It had a 50% potency compared to human IGF-I in a radioimmunoassay using an antiserum raised against human IGF-I, stimulated the incorporation of [3H]-thymidine into DNA in cultured chick embryo myoblasts with a half-maximum effective dose of 5 ng/ml and displaced [125I]-labelled human IGF-I and IGF-II from binding sites in microsomal membranes prepared from both the chicken liver and the lactating rabbit mammary gland in a dose dependent manner. 相似文献

Carbon-isotope values of bulk organic matter from high-altitude lakes on Mount Kenya and Mount Elgon, East Africa, were 10 to 14 per mil higher during glacial times than they are today. Compound-specific isotope analyses of leaf waxes and algal biomarkers show that organisms possessing CO2-concentrating mechanisms, including C4 grasses and freshwater algae, were primarily responsible for this large increase. Carbon limitation due to lower ambient CO2 partial pressures had a significant impact on the distribution of forest on the tropical mountains, in addition to climate. Nutrient supply may control muscle growth directly and indirectly through its influence on regulatory factors. The present study focuses on its effects on muscle insulin-like growth factors (IGF-I and -II) and myostatin (MSTN). Their mRNA levels were quantified by real time RT-PCR in pectoralis major (PM) and sartorius (SART) muscles from broiler chickens submitted to different feeding regimens (fed or fasted for 48 h) between hatch and 2 days of age and at 4 weeks of age. In the PM of 4 weeks old broilers, mRNA levels were also evaluated after a 16 h-fast and a refeeding period (refed 24 or 48 h after a 48 h-fast). In the PM muscle, both IGF-I and MSTN mRNA levels increased between 0 and 2 days of age in the fed group, while they remained low in the unfed one. A comparable trend was observed in the SART, but with lesser amplitude. In both muscles of 4 weeks old chickens, a 48 h-fast induced a significant reduction in MSTN mRNA levels (20% of fed state). In the PM, this effect required more than 16 h of fasting to occur and was fully reversed by only 24h of refeeding. IGF-I mRNA levels also varied with nutritional state. They decreased significantly with fasting in the SART muscle. By contrast, IGF-II mRNA levels did not vary significantly. Our data shows for the first time that two major paracrine regulators of muscle growth, IGF-I and MSTN, are sensitive to nutrient supply in hatching chicks, and also that fasting reduced IGF-I and MSTN mRNA levels in muscles of older chickens. 相似文献

Earthquakes on dipping faults: the effects of broken symmetry 总被引:3,自引:0,他引:3
Dynamic simulations of earthquakes on dipping faults show asymmetric near-source ground motion caused by the asymmetric geometry of such faults. The ground motion from a thrust or reverse fault is larger than that of a normal fault by a factor of 2 or more, given identical initial stress magnitudes. The motion of the hanging wall is larger than that of the footwall in both thrust (reverse) and normal earthquakes. The asymmetry between normal and thrust (reverse) faults results from time-dependent normal stress caused by the interaction of the earthquake-generated stress field with Earth's free surface. The asymmetry between hanging wall and footwall results from the asymmetric mass and geometry on the two sides of the fault. 相似文献

The effects of a selective bradykinin 1 receptor antagonist, compound A, were evaluated in a canine model of acute inflammatory model of arthritis. Despite detection of the B1 receptor in canine type B synoviocytes using a fluorescent ligand, oral administration of compound A (9 and 27 mg/kg) did not improve weight bearing of dogs injected intra‐articularly with IL‐1β in a force plate analysis. Analysis of the synovial fluid of IL‐1β‐treated dogs indicated high levels of bradykinin postchallenge. Excellent exposure, coupled with evidence of the presence of the B1 receptor during an acute inflammatory model of pain, indicates an inability of the receptor to mediate inflammatory pain in canines. 相似文献
